Transmission fluid is necessary for both options and you may need to change it occasionally. Most auto mechanics recommend changing the fluid every 60,000 miles. For a manual, the manufacturer will make recommendations in the owners booklet. The clutch will also need service from wear and tear, usually after about 60,000 miles, as well. Being harsh on your clutch may wear it down much sooner.
